Design Notes
GLS-shRNA-2 template sequence starts with GG, which is for transcription efficiency. GC content is around 50%, its target site is not in the first 75 bases after AUG, which are for the RNAi efficiency. UU overhangs in 3 prime-end, which is for the stability of shRNA. The antisense strand of shRNA is reverse complement with the sequence of its target site

GLS-shRNA-2 template is designed from the sequence of the target site in Glutathione S-transferase mRNA of Phyllotreta striolata, which is 576-596 after AUG. shRNA contains a sense strand, a loop, and an antisense strand.
